Abstract In this study, succinoglycan was produced from whey and its rheological and structural characteristics were elucidated. Eight culture media were prepared with different ions for bioconversion of whey into succinoglycan. The formulation composed of deproteinized whey, monobasic potassium phosphate and magnesium sulfate allowed the production of 13.7 ± 0.43 g/L of succinoglycan. The apparent molar mass of succinoglycan was estimated to be 9.033 × 105 g/mol and the polydispersity index was 1.044, representing the homogeneity of the sample. Monosaccharide composition of glucose and galactose for the succinoglycan produced was 6.6:1.0. The 1H RMN analysis revealed the non-saccharide substituent content of 1.2%, 3.0% and 8.1% for acetyl, succinate and pyruvate, respectively. The rheological results showed that the apparent viscosity of the succinoglycan solutions was directly proportional to the concentration, and the solution showed pseudoplastic behavior. Dynamic frequency sweep tests identified that a concentration of 2.0% of succinoglycan is required for formation of the gel system. Temperature influenced the viscoelastic behavior of succinoglycan and revealed the melting point and reversibility of the gel. Whey was shown to be a promising carbon source for the production of succinoglycan with thickening potential and viscosity modifier.

Abstract The aim of this study was to evaluate the effect of incorporating inulin and oligosaccharides resulted from the enzymatic hydrolysis of the commercial succinoglycan, regarding the physicochemical properties, rheology, syneresis and probiotic viability of soy and rice-based fermented beverages added with Lactobacillus paracasei. Four formulations were prepared and supplemented with 3.5% of inulin; 0.5% of succinoglycan oligosaccharides; 3.0% of inulin and 0.5% of succinoglycan oligosaccharides, and evaluated for 28 days of storage under refrigeration at 5 ºC. The succinoglycan oligosaccharides presented an average degree of polymerization (DPn) of 8. All the formulations were characterized as pseudoplastic fluids. Regarding syneresis, the formulations with succinoglycan oligosaccharides and Mix exhibited a reduction of it and this was not observed in the formulation with inulin. Regarding the probiotic viability, all the formulations maintained the subsistence of Lactobacillus paracasei with counts above 108 CFU mL-1 up to the 28th storage day, however, only the incorporated prebiotic formulations presented sufficient numbers of viable cells after exposure to simulated gastrointestinal conditions. Therefore, this fermented beverage proved to be an innovative alternative of non-dairy products interesting for the food industry to attend the population’s aspirations for functional products.

RESUMO: O objetivo deste estudo foi desenvolver massa alimentícia fresca sem glúten com adição de farinha de casca de maracujá amarelo (FCM). Uma formulação controle composta de uma mistura de farinha de arroz e farinha de milho, e formulações com adição de 10 e 20% de FCM foram produzidas. As amostras foram analisadas quanto às características sensoriais, físico-químicas e tecnológicas. A adição de FCM às formulações aumentou o tempo de cozimento, a perda de sólidos solúveis e a absorção de água, bem como, modificou os parâmetros de cor. As formulações que apresentaram melhores resultados, quanto aos atributos sensoriais, foram o controle e a que continha 10% de adição de FCM, que apresentaram índice de aceitabilidade acima de 70%. A adição de 10% de FCM aumentou o teor de fibras (3,25%) e cinzas (2,15%) e reduziu a porcentagem de carboidratos (41,19%) e o valor energético (223,26 kcal/80g), enquanto o teor de proteínas e lipídios foi semelhante entre formulações. A análise de componentes principais demonstrou que a amostra controle se destacou por suas propriedades sensoriais, enquanto a adição de 10% de FCM promoveu as propriedades nutricionais. Portanto, o aumento do valor nutricional da massa fresca sem glúten produzida com adição de FCM pode ser considerada uma alternativa para atender uma demanda crescente do mercado que busca escolhas alimentares mais saudáveis.

ABSTRACT: The aim of this study was to develop a gluten-free fresh pasta formulation with addition of yellow passion fruit peel flour (PPF). A control formulation containing a mixture of rice flour and corn flour and formulations with 10 and 20% of PPF addition were produced. Sensory analysis, physicochemical characterization and technological properties were assessed. The PPF addition to gluten-free pasta increased cooking time, soluble solids loss and water absorption; it also modified the color of the formulations. The control formulation and the formulation containing 10% PPF had the best sensory parameters, leading to an acceptability index over 70%. The 10% PPF addition increased fiber (3.25%) and ashes (2.15%) content and also lowered percentage of carbohydrates (41.19%) and energy value (223.26 kcal/80g); content of proteins and lipids were similar between the formulations. Principal component analysis showed that the control formulation stands out due to its sensory properties while the formulation containing 10% PPF improved nutritional properties. Therefore, the increase in nutritional value of gluten-free fresh pasta due to the PPF addition can be considered an alternative to meet market demands for healthier food choices.

The biosynthesis of amylase and collagenase, produced by A. acidocaldarius and A. sendaiensis respectively, were studied, and different matrices evaluated for the microorganisms immobilization and enzymes production optimization, such as loofa sponge, alginate-sponge and alginate, followed by concentration through ultrafiltration. Using a wheat bran substrate, the amylase enzyme displayed enzymatic activity of 0.45 U mL-1 and optimum temperature and pH conditions of 75 °C and pH 3.0, respectively. Thermal stability was in the range of 55 to 60 °C. The apparent Km and Vmax were 3.2 mg mL-1 and 0.5 U mL-1, respectively. The production of collagenase by A. sendaiensis was carried out with potato dextrose broth substrate and the activity obtained was 7.2 U mL-1. For amylase, the best results were obtained from immobilization in loofa sponge and the use of ultrafiltration (0.67 U mL-1) and for the collagenase extract, from the free biomass and ultrafiltration (13.6 U mL-1). The use of an ultrafiltration system enabled an average increase of 54% in the activity of both enzymes. Therefore, Alicyclobacillus are capable of producing enzymes of industrial interest, with the possibility of economically viable application of the substrate, and the use of immobilization and ultrafiltration produced positive results.

As bactérias do gênero Alicyiclobacillusspp.formam esporos e se desenvolvem em meios ácidos, podendo causar deterioração em sucos cítricos. O Brasil é o maior exportador de suco de laranja concentrado do mundo e, assim, este gênero vem sendo estudado por causar alterações de odor e sabor. Vários estudos relatam diferentes meios de culturas empregados para a detecção e enumeração de Alicyclobacillusspp.Este estudo teve como objetivo avaliar a recuperação de esporos de Alicyclobacillusspp.nos meios ALI, BAT, K ágar e YSG, utilizando a metodologia indicada pela ABECitrus. Cinco inóculos diferentes foram utilizados, sendo dois de linhagens-referência e os outros três isolados de suco concentrado de laranja pasteurizado. Também foi verificada a recuperação das células após o enriquecimento em suco de laranja reconstituído. Foi inoculada uma população inicial de 6 log UFC/mL. Os meios ALI, BAT e YSG conseguiram recuperar esta população nos diferentes inóculos, não existindo diferenças significativas entre os resultados. Contudo, devido à facilidade do preparo, os meios ALI e YSG mostraram-se mais vantajosos quando comparados ao meio BAT. O meio K ágar apresentou recuperação inferior aos outros meios para todos os inóculos, porém, houve diferença significativa apenas para Alicyclobacillus acidocaldarius 0298T (3,66 log UFC/mL) e Alicyclobacillus pomorum-like CBMAI 0278 (4,11 log UFC/mL).

Bacteria of the genus Alicyiclobacillus spp. form spores and develop in acid media, leading to the spoilage of citrus juices. Brazil is the largest exporter of orange juice concentrate, and yet, it has been extensively studied due to changes in taste and smell. Several investigations have reported different culture media used to detect and enumerate Alicyiclobacillus spp. The objective of the present study was to evaluate the recovery of Alicyiclobacillus spp. spores grown in ALI, BAT, K agar and YSG media using the methodology suggested by ABECitrus. Five inocula were used, two from reference strains and three from pasteurized concentrated orange juice. Cell recovery after the enrichment in reconstituted orange juice was also analyzed. An initial population of 6 log CFU/mL was inoculated. ALI, BAT and YSG media were able to recover the initial population of all different inocula, with no significant differences between the results. When compared to BAT, however, the preparation of ALI and YSG media was simpler and had more advantages. The recovery with K agar medium was lower than the other media for all the tested inocula, with significant differences found for Alicyclobacillus acidocaldarius0298T (3.66 log CFU/mL) and Alicyclobacillus pomorum-like CBMAI 0278 (4.11 log CFU/mL).
